Summary

  1. 1.

    The fine structure of capillary endothelium of the subfornical organ (SFO) differs from that of brain capillaries with respect to the following characteristics; it is fenestrated and contains a large amount of pinocytotic vesicles; the inner surface is enlarged by typical microvilli and tongue-like folds.

  2. 2.

    The vessels are surrounded by a palisade of radially arranged processes of neural, glial and parenchymal cells which form a highly enlarged adventitial surface. Between the vascular wall and the parenchymal palisade extends a wide extracellular space containing fibrous connective tissue as well as an electron middle dense substance. The inner basement membrane (300–500 Å) covers the outer surface of the endothelium and the pericytes. A complex system of outer basement membranes (300–500 Å) is investing the surface of individual processes of the palisade, and by penetrating the space of individual processes it forms a iwdely expanded interphase between vascular wall and parenchyma.

    The arterioles of the SFO have a tufted endothelium with pinocytotic vesicles and filaments (75–95 Å). A continuous line of smooth muscle cells containing filaments of 50–65 Å in diameter differentiates arterioles from the capillaries. Except in dimension there is no difference regarding the basement membranes (500–800 Å) and the extracellular space.

    Many structural characteristics of the SFO capillaries seem to be in agreement with those of other specialized brain areas (neurohypophysis, epiphysis, area postrema etc.) lacking a classical blood-brain barrier. The essential features are the greatly enlarged inner and outer surface of the capillary wall with the complex labyrinth of the outer basement membranes on one hand, the fenestrated endothelium with the highly developed pinocytotic activity on the other hand. These characteristics are interpreted in the light of increased transport functions.
