Summary
The incorporation of 3H-uridine, 3H-histidine, 3H-glucose, and 3H-palmitic acid into RNA, proteins, polysaccharides, and lipids of the optic tectum, the valvulae cerebelli and the cerebellum of 125 zebrafish (Brachydanio rerio) was investigated autoradiographically after incorporation periods of 11 min to 16 days under normal conditions. Additionally, the incorporation of 3H-uridine and 3H-histidine was investigated following different motor stimulations.

3H-uridine, 3H-histidine, and 3H-palmitic acid are strongly incorporated into the corresponding macromolecular compounds of the optic tectum, whereas the cerebellum and the valvulae cerebelli show significantly decreased tracer incorporation.

3H-glucose, on the other hand, is especially incorporated into the cerebellum.

While 3H-uridine-labelled RNA after its neosynthesis mostly remains within the region of the perikarya, 3H-histidine-labelled proteins are translocated proximo-distally by axonal flow. While lipids are synthesized ubiquitously in the nervous tissue, polysaccharides are mostly stored in the nerve fibre endings.

Following a weak motor stimulation, 3H-uridine and 3H-histidine are increasingly incorporated into the three CNS-structures investigated; following a strong motor stimulation, there is a decrease of tracer incorporation into the optic tectum and the valvulae cerebelli, whereas the cerebellum shows a constant incorporation.

Animals inhibited in their motility show a significantly decreased incorporation of RNA- and protein precursors.
Zusammenfassung
An 125 Zebrafischen (Brachydanio rerio) wurde der Einbau von RNS-, Protein-, Polysaccharid und Lipoid-Tracern unter Normalbedingungen, sowie der Einbau von 3H-Uridin und 3H-Histidin in Abhängigkeit von einer adäquaten Motilitätsstimulation im Tectum opticum, in den Valvulae cerebelli und im Cerebellum autoradiographisch untersucht.

3H-Uridin, 3H-Histidin und 3H-Palmitinsäure werden im ZNS besonders stark in die entsprechenden makromolekularen Verbindungen des Tectum opticum eingebaut, während das Cerebellum und die Valvulae einen geringeren Tracer-Einbau aufweisen.

3H-Glucose wird dagegen vornehmlich in das Cerebellum inkorporiert.

Während die 3H-Uridin-markierte RNS nach ihrer Synthese vornehmlich im Perikaryenbereich des Nervengewebes verbleibt, werden mit 3H-Histidin-markierte Proteine proximo-distal mit dem axonalen Transport transloziert. Die Lipoid-Synthese erfolgt im Nervengewebe ubiquitär, Polysaccharide werden vornehmlich in den Nervenfaser-Endformationen gespeichert.

Nach einer schwachen motorischen Belastung werden 3H-Uridin und 3H-Histidin mit regionsspezifischen Unterschieden verstärkt in die drei untersuchten Strukturen des ZNS eingebaut; nach einer starken motorischen Stimulation findet hingegen ein verminderter Tracer-Einbau in das Tectum opticum und die Valvulae statt, während das Cerebellum eine weitgehend konstant bleibende Tracer-Inkorporation aufweist.

Motorisch inaktivierte Tiere zeigen sowohl nach kurzen als auch längeren Versuchszeiten eine signifikant verminderte Tracer-Inkorporation.
