The composition of the hemolymph of the crayfish Orconectes limosus was analysed with respect to seasonal variations of its constituents and to the influence of moulting, sex, size and injury. Freezing point depression, sodium, potassium, calcium, magnesium, chloride, acid-soluble phosphate, total phosphorus, lactic acid, copper, protein, non-protein nitrogen, urea, acid-soluble glucose and hexose, number of blood cells and pH were analysed for a 14 months period using 450 animals.

Protein is the most important buffer substance in the hemolymph. Only 18% of the buffer capacity is removed by dialysis. The hemolymph contains about 0.5% copper-free protein. The copper content of crayfish hemocyanin was 0.176 mg copper per g protein. Ultrafiltration experiments showed that sodium, potassium, chloride, glucose and urea pass freely into the filtrate. About one third of the amounts of total phosphorus, non-protein nitrogen and acid-soluble hexose are retained. 18% of both calcium and magnesium appear to be bound by protein.

Most of the constituents analysed showed the same annual pattern of variation: the greatest concentrations were found during the winter months, followed by a sharp decline in the spring, a recovery to medium summer levels before the beginning of the moulting period and a subsequent rise to higher winter values. The amplitude of variation was about ±10% around the mean for the freezing point depression, chloride and sodium, but larger for most other parameters. From the close correspondence between changes in freezing point depression and temperature of the ambient water it was deduced that temperature is a major factor in determining the overall concentration of the hemolymph.

Changes in hemolymph composition in connection with moulting were studied in June and July using 160 animals. During premoult the concentrations of all constituents rise above the intermoult level (about 10% in the case of the inorganic ions). With ecdysis there is a sharp decline in concentrations. Sodium, chloride and potassium decrease by 30% to about 80% of the intermoult values. Total phosphorus, magnesium, non-protein nitrogen and protein reach intermoult levels, while glucose and acid-soluble phosphate decline much farther. Calcium values remain above the intermoult level for about one week following moult. Urea is exceptional as its concentration rises from late premoult through ecdysis and postmoult. The amount of water taken up during the moulting process is calculated to be equivalent to at least 12% of the body weight.

Sexual variation in hemolymph composition seems to be very limited. Females have higher glucose and protein levels and smaller freezing point depressions during winter and higher calcium concentrations during the summer months than males.

Correlation analysis revealed that larger animals tend to have higher potassium values, lower freezing point depressions and lower levels of most of the organic constituents than smaller ones. There is no significant correlation between size and sodium, chloride, calcium or magnesium concentrations.

The changes in hemolymph composition which occured with oviposition and after injury were qualitatively similar to those observed after ecdysis, although the decline of concentrations was less evident.

Van Harreveld's saline seems to be a suitable physiological solution for the animal under consideration. However, suggestions have been made to replace the original bicarbonate buffer by an organic buffer system.

Die Zusammensetzung der Hämolymphe des Flußkrebses Orconectes limosus und die Veränderung einzelner Bestandteile im Laufe des Jahres wurden ebenso wie der Einfluß der Häutung und die Einflüsse von Verwundung, Geschlecht und Körpergröße untersucht. Folgende Parameter wurden über einen Zeitraum von 14 Monaten an 450 Einzeltieren bestimmt: Gefrierpunktserniedrigung, Natrium, Kalium, Calcium, Magnesium, Chlorid, säurelösliches Phosphat, Milchsäure, Gesamtphosphor, Kupfer, Protein, Reststickstoff, Harnstoff, säurelösliche Glukose und Hexose, Zahl der Blutzellen und pH.

Das Blutprotein ist die wichtigste Puffersubstanz. Durch Dialyse sind nur 18% der Pufferkapazität der Hämolymphe zu entfernen. Das Blut enthält im Mittel 0,5% kupferfreies Protein. Der Kupfergehalt des Hämocyanins wurde zu 0,176 mg Kupfer pro g Protein bestimmt. Ultrafiltrationsversuche ergaben, daß Natrium, Kalium, Chlorid, Glukose und Harnstoff vollständig, Gesamtphosphor, Reststickstoff und säurelösliche Hexose zu etwa zwei Dritteln ultrafiltrierbar sind. Calcium und Magnesium sind zu 18% proteingebunden.

Den Veränderungen, die im Laufe des Jahres bei fast allen Parametern in gleichsinniger Weise zu finden waren, liegt folgendes Schema zugrunde: Von einem relativ hohen Winterniveau fallen die Werte im Frühjahr stark ab, steigen dann noch vor Beginn der Häutungen auf ein mittleres Sommerniveau an, von dem es im Herbst wieder zu höheren Werten geht. Die Amplitude der Veränderungen beträgt bei der Gefrierpunktserniedrigung, bei Natrium und Chlorid ±10% um das Jahresmittel, während sie bei den meisten anderen Parametern größer ist. Aus dem gleichsinnigen Verlauf der Kurven für die Wassertemperatur und die Gefrierpunktserniedrigung des Blutes wurde geschlossen, daß der jahreszeitliche Temperaturwechsel die Veränderungen in der Konzentration der einzelnen Blutbestandteile bedingt.

Veränderungen im Zusammenhang mit der Häutung wurden in den Monaten Juni und Juli an 160 Tieren ausgesuchter Häutungsstadien untersucht. Vor der Häutung steigen alle Blutwerte über das Zwischenhäutungsniveau an (10% im Fall der anorganischen Ionen). Mit der Häutung sinken sie stark ab, wobei Natrium, Kalium und Chlorid bis auf etwa 80% der Zwischenhäutungswerte fallen. Gesamtphosphor, Magnesium, Reststickstoff und Protein gehen etwa auf die Ausgangswerte zurück, während Glukose und säurelösliches Phosphat stärker abfallen. Harnstoff steigt während der späten Vorhäutung und der Nachhäutung ständig an. Die Calciumwerte bleiben länger als 1 Woche nach der Häutung über dem Zwischenhäutungsniveau. Bei der Häutung wird mindestens die 12% des Körpergewichts äquivalente Wassermenge aufgenommen, wodurch das Blut um etwa 30% verdünnt wird.

Es scheinen nur geringe Geschlechtsunterschiede in der Blutzusammensetzung vorzuliegen. Weibchen zeigen im Winter höhere Protein- und Glukosewerte und eine niedrigere Gefrierpunktserniedrigung als Männchen. Die Männchen haben im Sommer geringere Calciumwerte als die Weibchen.

Die Korrelationsanalyse ergab, daß größere Tiere im Mittel höhere Kaliumwerte, kleinere Gefrierpunktserniedrigungen und geringere Konzentrationen der meisten organischen Blutbestandteile haben. Zwischen Körpergröße einerseits und den Natrium-, Chlorid-, Calcium- und Magnesiumkonzentrationen andererseits besteht kein signifikanter Zusammenhang.

Die bei der Eiablage und nach einer Verwundung festgestellten Veränderungen in der Zusammensetzung des Blutes sind qualitativ ähnlich wie die bei der Häutung beschriebenen, allerdings sind die Konzentrationsänderungen geringer.

Eine Abänderung der van des dort verwendeten Bikarbonatpuffers durch ein organisches Puffersystem Harreveldschen Blutersatzflüssigkeit erscheint nicht notwendig, aber der Ersatzwird vorgeschlagen.
