Summary
In order to evaluate prolactin secretion by autotransplanted pituitaries in eels Anguilla anguilla, some biological phenomena under prolactin control were collected: the histological structure of the kidney, plasma sodium, and calcium levels, and the water content of muscle.
Previously, we showed with A. M. Lemoine that the epithelium (cell height and nuclear size) in kidney tubules was affected after hypophysectomy and partly restored with ovine prolactin treatment. A similar kidney activation is observed in an intact eel in which prolactin cells were spontaneously overstimulated.
A comparison of kidneys in intact and hypophysectomized eels, and in eels which have had an autotransplanted pituitary for 2 months confirms that hypophysectomy leads mainly to a decrease of nuclear surfaces in the various segments of kidney tubules after 2 months; in grafted eels, the epithelial cell height remains normal, but the nuclear surfaces are intermediate between those of intact and operated animals; that is, nuclear atrophy is partly prevented by the graft.
Prolactin plays an important role in the water and electrolyte metabolism of eels. After hypophysectomy a 36% decrease of blood sodium level is noted as well as a significant blood calcium diminution, and muscle water content is increased. In grafted eels blood sodium decrease is only 8.45%, calcium level is normal, and water content, although below that of hypophysectomized eels, remains higher than in the controls.
Data collected on kidney structure, plasma sodium, and muscle water content show that a graft is unable to completely prevent the effect of hypophysectomy. This suggests a deficit of prolactin secretion despite the histological picture of a high stimulation of the prolactin cells in the transplant.
The hypothesis of a PIF and a PRF (hypothalamic factors of prolactin inhibition and release) is discussed and similarities with ectopic pituitary in birds are noted.
Résumé
Afin d'évaluer la production de prolactine par l'hypophyse de l'Anguille après déconnexion hypothalamique, nous avons étudié divers paramètres biologiques contrôlés par cette hormone: structure histologique du rein, sodium et calcium plasmatiques et teneur en eau du muscle.
Ayant montré, avec A. M. Lemoine, que l'épithélium des tubes rénaux est affecté par l'hypophysectomie et en partie préservé par l'apport de prolactine ovine, nous avons comparé le rein d'Anguilles intactes, hypophysectomisées et autogreffées depuis 2 mois. Chez un témoin, aux cellules á prolactine extrêmement actives, la néoformation de néphrons est très stimulée. Nous avons vérifié que 2 mois après hypophysectomie l'atrophie porte principalement sur les surfaces nucléaires, étant moins marquée sur la hauteur des épithéliums. L'atrophie est totalement compensée par l'autogreffe pour la hauteur des cellules, mais en partie seulement pour les aires nucléaires des divers segments tubulaires.
La prolactine joue un rôle important dans l'équilibre hydrominéral de l'Anguille. Après hypophysectomie, nous avons confirmé la chute du sodium plasmatique qui atteint 36% et une baisse très significative du calcium, tandis que la teneur en eau du muscle s'accroît. Chez les Anguilles greffées, la chute du sodium est fortement réduite (8,5% seulement), celle du calcium n'apparaît plus, mais la teneur en eau du muscle est intermédiaire entre celle des intactes et des opérées.
Pour les paramètres étudiés, sauf pour la calcémie, la greffe est donc incapable de compenser complètement les effets de l'hypophysectomie, et l'hypothèse d'un déficit de la sécrétion globale de prolactine est envisagée, en dépit de l'image de forte stimulation des cellules à prolactine du greffon. La présence hypothétique d'un PIF et d'un PRF (facteurs hypothalamiques inhibant ou stimulant la décharge de prolactine hypophysaire) est discutée et la similitude avec l'hypophyse ectopique des Oiseaux est soulignée.
