Chromatography by silica-gel chromatobars

Prospects of its application in the field of forensic toxicology

Abstract

A new type of Chromatographic column of silica gel 20 mm in diameter and 180 mm long has been investigated in the field of forensic analytical toxicology.

In order to evaluate the usefulness of chromatobars in isolating organic compounds from biological extracts, sodium phenyl-ethyl-barbiturate, atropine, morphine and strychnine were added to the aqueous extract obtained with StasOtto's isolation method.

Afterwards the residues of the acidic ether extract, alkaline ether extract and alkaline chloroform extract, were chromatographed on chromatobar.

This chromatographic device seems able to avoid the drawbacks and limitations of paper and thin-layer chromatography, mainly from the preparative aspect. The method is particularly advantageous in analysing the whole extract obtained with the Stas-Otto process, without bringing about its partition, especially when slight traces of toxic substances are present.

Zusammenfassung

Ein neuer Typ der Chromatographie-Säule aus Silicagel (20 mm Durchmesser und 180 mm Länge) wird in die forensisch-toxikologische Analyse eingeführt. Zur Erprobung der Brauchbarkeit des „Chromato-barrens” für die Isolierung organischer Verbindungen aus biologischem Material wurden Phenylaethylbarbitursaures Natrium, Atropin, Morphin und Strychnin (je 1 mg) zu dem nach Stas-Otto gewonnenen wäßrigen Extrakt (aus 300 g Gewebe von Hirn, Nieren und Leber) zugesetzt. Danach wurden die Rückstände des sauren Ätherextraktes, des alkalischen Ätherextraktes und des alkalischen Chloroformextraktes am Chromato-barren chromatographiert. Diese Methode scheint geeignet, Nachteile und Begrenzungen der Papier- und Dünnschichtchromatographie — besonders aus präparativer Sicht — zu vermeiden. Die Methode ist besonders vorteilhaft bei der Aufarbeitung des Stas-Otto-Gesamtextraktes ohne Zwang zur Aufteilung, insbesondere wenn nur Spuren toxischer Substanzen vorhanden sind.
