Localization of RNA synthesis sites in the 1B-3C region of the Drosophila melanogaster X Chromosome

Abstract

RNA synthesis following 3H-uridine pulse labeling has been studied autoradiographically in the 1B-3C region of D. melanogaster salivary gland X-chromosome. To improve the precision of RNA synthesis site localization in bands and interbands a micromanipulator technique of chromosome stretching was used. No significant RNA synthesis was detected in the interbands. RNA synthesis was found to occur not only in puffs but also in bands whose puffing was not apparent. In 5 genetically studied segments of the 1B-3C region where 56 complementation groups have been identified (Lefevre, 1974; Rayle, 1972; Gvozdev et al., 1975; Judd et al., 1974; Lefevre and Green, 1972), at least 21 transcription sites are detected.
