Summary
Human peripheral blood and cultured human lymphocytes undergoing the first mitosis were exposed to ultrasound in order to find out whether ultrasound can induce chromosome aberrations.
Special vessels which prevented reflections were used for insonation. The experiments were carried out by a method avoiding any heating of the sonicated medium. During the investigations continuous ultrasound up to an intensity of 0.2 W/cm2 at a frequency of 2 MHz and up to an intensity of 35 W/cm2 at a frequency of 870 kHz was applied. In addition, pulsed sound of an average intensity of 0.02 W/cm2 at a frequency of 2 MHz was used (pulse sequence 2000/sec, pulse length 1 μsec). Afterwards chromosome cultures from lymphocytes were prepared and examined for numerical and structural aberrations. In spite of the high intensities applied no damaging effect on the chromosomes or the spindle were found. Mitotic activity was not affected by the insonation.
Zusammenfassung
Um festzustellen, ob Ultraschall Chromosomenaberrationen zu induzieren vermag, wurden menschliches Venenblut und ankultivierte menschliche Lymphocyten in Teilung mit Ultraschall behandelt.
Bei der Beschallung wurden Spezialgefäße benutzt, die Reflexionen ausschließen. Die Versuchsanordnung wurde jeweils so gewählt, daß eine Erhitzung des beschallten Mediums möglichst vermieden wurde. Zur Anwendung kam bei den Versuchen Dauerschall bis zu einer Intensität von 0,2 W/cm2 bei einer Frequenz von 2 MHz und bis zu einer Intensität von 35 W/cm2 bei einer Frequenz von 870 kHz. Außerdem wurde Impulsschall einer mittleren Intensität von 0,02 W/cm2 bei einer Frequenz von 2MHz benutzt (Pulsfolge 2000/sec, Pulsdauer 1 μsec). Anschließend wurden die Chromosomen aus den Lymphocyten dargestellt und auf numerische und strukturelle Aberrationen untersucht. Trotz der hohen angewendeten Intensitäten konnte weder eine chromosomenschädigende Wirkung noch eine Schädigung des Spindelapparates der Mitose festgestellt werden. Die Zellteilungsaktivität blieb unter den Beschallungen ebenfalls unbeeinflußt.
