Comparative studies on the in vivo distribution pattern of dibromodulcitol and diepoxydulcitol

Summary

Comparative studies of the in vivo distribution pattern of two cytostatic compounds — 1.6-dibromo-dulcitol and 1.2–5.6-diepoxy-dulcitol (a derivative developed in alkaline media) — were carried out in mice and rats by using compounds labelled with tritium in position 1. In the course of these investigations some of the biological transport parameters of 1.6-dibromo-dulcitol and of 1.2–5.6-diepoxy-dulcitol were found to be similar while others differed. Both compounds are characterized by different tissue uptake and accumulation rates. For 1.6-dibromo-dulcitol high specific concentration values exceeding the average could be recorded in brain tissue, shortly after treatment whereas following 1.2–5.6-diepoxy-dulcitol treatment peak values were found in tissues of the gastro-instestinal tract. The accumulation of 1.2–5.6-diepoxy-dulcitol was 2–3 times higher than that of 1.6-di-bromo-dulcitol.

Zusammenfassung

Es wurden die „in vivo“ Verteilungseigenschaften des cytostatischen 1,6-Dibromdulcit und des sich daraus in alkalischem Medium entwickelnden 1,2-5,6-Diepoxydulcit mit Hilfe von in Position eins mit Tritium markierten Verbindungen an Mäusen und Ratten untersucht. Den Prüfungen gemäß konnten im biologischen Transport des 1,6-Dibromund des 1,2–5,6-Dianhydro-dulcit neben ähnlichen Tendenzen auch einzelne Unterschiede beobachtet werden. Die Gewebeaufnahme und die Kumulationsfähigkeit waren in beiden Verbindungen unterschiedlich voneinander. Kurz nach der Behandlung zeigte das 1,6-Dibromdulcit im Gehirngewebe, das 1,2-5,6-Dianhydrodulcit jedoch in Gewebe des Verdauungssystems vom Durchschnitt abweichende hohe spezifische Konzentrationswerte. Die Kumulation des Diepoxydulcit war 2–3 mal stärker als die des DBD.
