Summary
Within the first 6 hrs of treatment of human lymphocytes with Phenylbutazone (Phb) the mitotic index is staggering very much. This is interpreted as expression of an intensive biochemical activity in the cells at the end of S-Phase and G2-Phase.
In cultures for comparison, to which 2 hrs before preparation 0.2 ml of Colcemid-solution had been added, the numerous proportion of the single mitotic stages was evaluated. By it the “Mitotic stages-Index” was defined.
In the cultures treated with Phb the Mitotic stages-Indices have been evaluated too. By them the different moments of the cell cycle could be determined, in which Phb provides cells from entering mitosis and inhibits the passage from prophase to metaphase. From this was concluded that the succession of mitotic stages as well as the separation of the centromeres are independent of a developped mitotic apparatus as soon as cell division has started. Obviously the separation of the centromers can be influenced by Phb before it is decided whether the cells enter mitosis or not.
The probable mechanism of reaction of Phenylbutazone and the possibilities which result from the methods used, to test and develop cytostatic agents, are discussed.
Zusammenfassung
Innerhalb der ersten 6 Std der Behandlung menschlicher Lymphocyten mit Phenylbutazon (Phb) ist der Mitoseindex starken Schwankungen unterworfen. Dies wird als Ausdruck einer intensiven biochemischen Aktivität der Zellen gegen Ende der S-Phase und während der G2-Phase interpretiert.
In Vergleichskulturen, welchen 2 Std vor der Aufbereitung 0,2 ml Colcemid-Lösung zugesetzt wurden, wurde das zahlenmäßige Verhältnis der einzelnen Mitosestadien zueinander bestimmt und damit der «Mitosestadien-Index» definiert.
An Hand der in den mit Phb behandelten Kulturen ermittelten Mitosestadien-Indices wurden die verschiedenen Zeitpunkte im Zellcyclus bestimmt, an welchen Phb den Eintritt in die Zellteilung und den Übergang von der Prophase in die Metaphase hemmt und die Zentromertrennung beeinflußt. Daraus wurde geschlossen, daß die Aufeinanderfolge der Mitosestadien einschließlich der Zentromertrennung von einem ausgebildeten Spindelapparat unabhängig ist, sobald die Zellteilung einmal begonnen hat. Die Zentromertrennung selbst ist ganz offensichtlich durch Phb bereits beeinflußbar, noch bevor die Entscheidung darüber gefallen ist, ob die Zellen in die Mitose eintreten order nicht.
Der wahrscheinliche Wirkungsmechanismus von Phenylbutazon und die sich aus der angewandten Methode ergebenden Möglichkeiten zur Untersuchung und Entwicklung cytostatisch wirksamer Pharmaka werden diskutiert.
