Stimulating effect of serum from tumor-bearing mice on lymphocyte response to mitogenic stimulation associated with protein increase in serum

Summary

The influence of serum from mice bearing large tumors on the 3H-thymidine uptake of normal mouse spleen cells in culture following PHA and Con A stimulation was investigated.

Some sera produced a substantial increase in 3H-thymidine uptake after mitogenic stimulation, for 0.1%–5% dilution, compared with control serum or serum taken from other tumor-bearing mice.

Determination of the amount of proteins present and polyacrylamide gel electrophoretic analysis revealed higher protein values in these sera than in control serum. Sera from tumor-bearing mice that induced a decrease in 3H-thymidine uptake also contained lower amounts of protein than control serum.

Some sera, which were heated to 56° C, kept a lower stimulating action at 0.1%–1% dilution. At 5% dilution, a reverse action was observed. Sera that had a stimulating action showed a depressive action when compared with control serum heated to 56° C.

Electrophoretic analysis of serum heated to 56° C showed significant differences, principally for β-lipoproteins and γ-globulin fractions.
