Summary
A very sensitive radioisotope method, using 125I-labelled serum albumin as substrate, is proposed for the detection of very low enzymatic activities released from insoluble matrix to eluate, storage media and washings after an immobilization procedure. In the case of papain, proteolytic activities corresponding to about 0.1μg of enzyme per ml can be detected. In contrast, simple immunochemical techniques did not give any increase in sensitivity in papain determination compared with the conventional modified Anson method.
