We report here our studies of the morphological signs of the cerebroprotective influence of the polyprenol agent Ropren on astrocytes in the paranigral nucleus of the midbrain, located outside a focus of forebrain ischemia, in rats. Mature rats (n = 8) were subjected to forebrain ischemia under general anesthesia by bilateral occlusion of the common carotid arteries. After surgery, four of the animals received daily i.p. injections of the cerebroprotector Ropren at dose of 11.6 mg/kg. Controls consisted of sham-operated animals. Animals were decapitated on experimental day 7. Astrocytes in the anterior ventral part of the paranigral nucleus of the midbrain were studied immunohistochemically by detecting glial fibrillary acidic protein. The numbers of astrocytes, the cross-sectional areas of their bodies, the lengths of their main processes, and the distances between their bodies and capillary walls were measured. Ropren was found to suppress the hypertrophic reaction of astrocytes and their migration towards vessel walls, and appeared to stimulate the multiplication of these cells.
