Abstract
Black cohosh ( Actaea racemosa L., syn. Cimicifuga racemosa (L.) NUTTALL) native to eastern North America, is member of the plant family Ranunculaceae. The perennial plants are growing in deciduous woodlands. Products of dried plant rhizomes (Cimicifugae racemosae rhizoma) and lateral roots (Radix Cimicifugae racemosae) mostly from wild-grafted plants are used in phytotherapy. For field cultivation homogenous plant material is required, but seed progenies are genetically and phenotypically heterogeneous. Somatic embryogenesis was applied for vegetative mass propagation of selected, high-value plants. As explants stamens from immature flower buds were used. The Murashige and Skoog (15:473–479, 1962) medium was modified for stages as embryogenic mass initiation, maintenance and propagation of embryogenic mass, embryo maturation, embryo conversion, and plant development. Embryogenic mass was induced by application of 1 mg/l 2,4-D. Embryo development and maturation was supported by using a hormone-free medium supplemented with 1% activated charcoal. Application of 0.2 mg/l 6-benzyladenine improved bipolar conversion of embryos. In field, the plants produced via somatic embryogenesis were homogenous in phenotype.
