Abstract

Two types of general methods are detailed in this chapter. The first involves formation of a Holliday junction substrate that can be used to measure both helicase-dependent Holliday junction branch migration and Holliday junction resolution driven by Ruv proteins from any bacterial species, whether mesophilic or thermophilic. The second involves the purification of Ruv proteins from the thermophile, Thermotoga maritime, by procedures applicable to the purification of thermostable proteins from a variety of thermophilic genera. The specific assays detail the measurement of branch migration and resolution of the Holliday junction substrate by T. Maritime Ruv proteins.
