Definition
Histaminic agonists and antagonists are drugs that mimic or antagonize the actions of histamine at one of its four G-protein-coupled receptors (H1–H4).
Pharmacological Properties
Histamine is a biogenic amine that is widely distributed in the body and is involved in a number of physiological and pathophysiological functions (Hill et al. 1997; Parsons and Ganellin 2006). These include allergic inflammation, cardiovascular regulation, gastric acid secretion, and neuromodulatory functions within the central nervous system (CNS). Histamine is produced by decarboxylation of the amino acid l-histidine and is metabolized by histamine NÏ„-methyltransferase (HMT) and diaminoxidases. In the case of its neurotransmitter role within the CNS, histamine-containing neurons are characterized by a high level of histidine decarboxylase activity and removal of synaptic histamine is mainly via metabolism (as a consequence of HMT activity)....
