Molecular and general genetics of ectomycorrhizal fungi

Abstract

The variability observed in ectomycorrhizal (EM) fungi provides a substantial genetic resource and the potential to enhance forest productivity and greening of waste and degraded lands. Classical and molecular genetics are being developed to obtain genetic information for the selection of improved EM fungal strains. The understanding of sexual and parasexual reproduction, post meiotic nuclear behaviour, mating types and vegetative incompatibility mechanism is essential for the manipulation of EM fungi. The genetic traits which contribute to symbiosis and plant function are being understood by combining natural variability with classical and molecular genetic manipulations. The developments in the manipulation of genomes of EM fungi depend upon the efficient methods for gene cloning and DNA transformation. The successful attempts in transforming EM fungi have been discussed. The genetic variability of EM fungi can be assessed by DNA fingerprinting. This approach would be useful for the evaluation of variability and exchange of genetic information in natural populations, the identification of the species and isolates by DNA polymorphism and tracking the environmental fate of the introduced fungi to find out their survival, growth and dissemination within the soil. The possibilities shown in developing DNA probes based on nuclear DNA and mitochondrial DNA have been discussed.
