Abstract

An increase in plasma HDL cholesterol (C) after incubation of whole blood or plasma at 37°C has been previously described. To differentiate between the influence of cells and of plasmatic factors on HDL-C, we compared changes in HDL-C during 4 h at 37°C in whole blood and plasma samples from 31 healthy subjects. HDL were separated by phosphotungstate/MgCl2 precipitation. Total, free, and esterified plasma and lipoprotein cholesterol were enzymatically determined in triplicate. Results: i) Increase in HDL-C was not different in whole blood and plasma, despite significant cholesterol transport out of the cells. ii) In 55 % of the blood samples increase in HDL-C was more pronounced than increase in total cholesterol. Therefore, HDL had taken up VLDL/LDL cholesterol, also in the presence of blood cells. Hi) Differences were found mainly in the distribution of newly formed cholesteryl esters. In subgroups two extreme patterns were observed: all newly formed cholesteryl esters were either found in the HDL fraction or in the VLDL/LDL fraction. Moreover, net transfer of VLDL/LDL cholesteryl esters to the HDL was demonstrable in a further subgroup. It is suggested that plasmatic factors, such as cholesteryl ester transfer, may be of greater importance in the regulation of HDL-C than cholesterol transport out of the cells.

Zusammenfassung

In vorausgegangenen Untersuchungen wurde nach Inkubation von Vollblut bei 37°C ein Anstieg von High density lipoproteins(HDL)-Cholesterin (C) im Plasma beobachtet. Um zwischen dem Einfluß von Blutzellen und von Plasmafaktoren auf den HDL-C-Spiegel zu unterscheiden, wurden Vollblut- und Plasmaproben von 31 gesunden Spendern parallel über 4 Stunden bei 37°C inkubiert und die Änderungen im Plasma- und Lipoproteincholesterin verglichen. Cholesterin (gesamt, frei und verestert) wurde enzymatisch in Dreifachbestimmungen gemessen, in der HDL-Fraktion nach Fällung mit Phosphorwolframat/MgCl2. Ergebnisse:

    Bei ca. 50 % der Blutproben war der Anstieg im HDL-C größer als der im Gesamtcholesterin. Folglich hatten die HDL auch in Gegenwart von Blutzellen Cholesterin von den Very low density lipoproteins (VLDL)/Low density lipoproteins (LDL) aufgenommen.

    Der Anstieg im HDL-C unterschied sich zwischen Vollblut- und Plasmaproben nicht, obgleich ein signifikanter Cholesterintransport aus den Blutzellen nachzuweisen war.

    Subgruppen der Probanden zeigten erhebliche Differenzen in der Verteilung der Cholesterinester zwischen den Plasmalipoproteinen: Neugebildete Cholesterinester waren entweder nur in den HDL oder nur in den VLDL/LDL wiederzufinden. In einer weiteren Subgruppe war ein Nettotransfer von Cholesterinestern von den VLDL/LDL zu den HDL zu beobachten. Die Befunde sprechen dafür, daß plasmatische Faktoren, wie der Cholesterinestertransfer, den HDL-C-Spiegel möglicherweise stärker beeinflussen als der Cholesterintransport aus Zellen.
