Abstract
Glycan microarray allows for direct measurement and characterization of the carbohydrate–protein interactions and only a very small amount of sugar is required. In addition, it may not only effectively mimic the presentation of glycans on the cell membrane but also yields high affinity and specificity to exhibit the multivalent interaction with receptor. With these synthetic carbohydrates in hand, the structure of the synthetic product was verified by 1D and 2D NMR analysis. Besides, we also used the RM2 antibody for binding analysis on a glycan array to confirm our synthetic structure. Furthermore, we use carbohydrate microarray to further characterize the binding specificities of these RM2 and its truncated analogs with monoclonal antibody RM2.
