Abstract

This chapter will focus on biological rhythms of gastrointestinal (GI) function insofar as they influence the pharmacokinetics, pharmacodynamic and toxicological behavior of oral and, in some instances, parenterally administered medications. Rhythms around the circadian (~24 h) and ultradian ( <22 h) time domains will be emphasized; infradian ( >28 h, to include monthly and yearly rhythms) rhythms will not be discussed simply because there is no published evidence of their influence, or even existence, in human gastrointestinal function. Human data will be highlighted; animal data will be included only if supportive of a human observation or in support of a particular theoretical view proposed by the authors. In addition, rhythms of motor, secretory or absorptive function of the hollow-viscus GI organs will receive greater attention; those of the liver — important as this organ is in drug metabolism — and pancreas are beyond the intended scope of this chapter and are dealt with in more detail elsewhere (Belanger and Labrecque 1992; Mejean et al. 1992).
