Abstract
Niemann-Pick disease (NPD) is a rare hereditary disorder of lipid metabolism which is characterized by deposition of sphingomyelin in endothelial, mesenchymal and parenchymal cells of almost every organ and tissue. Its clinical manifestations appear in most instances during the first year of life. The symptomatology consists of hepatosplenomegaly and neurological disturbances with retardation of somatic and mental development. Affected children usually do not reach their third year of life. In exceptional cases the first symptoms appear later in life and progression of the disease is slower. The underlying error of metabolism is still not known with certainty. It appears that the metabolic lesion in the classic infantile form of NPD is attributable to a deficiency or lack of activity of the enzyme which catalyzes the cleavage of sphingomyelin (Brady et al. 1966).
