Analysis of Intracellular Antigens by Flow Cytometry: Methods for Cell Permeabilization to Antibodies

Abstract

Intracellular molecules have been detected for many years using antibodies and other specific ligands conjugated to fluorescent markers. Cells were prepared by established methods of histochemistry and cytochemistry and were analyzed by fluorescence microscopy. By microscopy not only the presence of an intracellular antigen can be identified but also its localization within the cell. Quantitative analysis, however, suffers from the limitations of microscopy in general, namely (1) the relatively small number of cells of a large population that are analyzed, (2) the slowness and subjectivity of data collection and (3) the restricted sensitivity of the human eye to low-intensity fluorescence.
