Abstract

Since the first reports on the use of site directed mutagenesis in 19821 protein engineering or — when rationally aimed — protein design has been recognized as a promising and fascinating field of research in many countries. In Japan (PERI) and the U.S.A. (CARB) research institutes have been founded with the focus on protein design. More and more research institutes in the United States, Canada, Japan and Europe have been starting broad research projects on protein design (UK: SERC, W. Germany: CAPE).

Nr. 1a of Fig. 2 describes the task of modelling an unknown 3D-structure of an existing protein out of a known structure of a homologuous protein whereas Nr. 4 and 5 are standing for the task of predicting 3D-structure changes after exchange of amino acids in a protein of known 3D-structure. The methods used are rather similar.
