Zusammenfassung
5% aller Mamma- und Ovarialkarzinomerkrankungen haben eine genetische Ursache. Die beiden Hauptgene in diesem Zusammenhang sind die Gene BRCA1 und BRCA2. Berücksichtigt man eine Brustkrebsinzidenz von 45.000 Neuerkrankungen pro Jahr, so sind schätzungsweise jährlich in Deutschland 1000 Fälle auf BRCA-1- und 800 Fälle auf BRCA-2-Mutationen zurückzuführen. Bei 8300 Ovarialkarzinomneuerkrankungen in Deutschland basieren etwa 370 Ovarialkarzinomfälle jährlich auf Mutationen im BRCA-1-Gen. In mindestens 90% aller Familien mit dem gehäuften Auftreten von Mamma- und Ovarialkarzinomen und in 40% aller Familien mit einem gehäuften Auftreten von Mammakarzinomen sind Mutationen des BRCA-1-Gens direkt für die Tumorentstehung verantwortlich. Das kumulative Risiko für BRCA-1-Gendefektträge- rinnen, bis zum 80. Lebensjahr an einem Mammakarzinom zu erkranken, ist 80%. Das Risiko dieser Patientinnen, bis zum 80. Lebensjahr an einem Ovarialkarzinom zu erkranken, liegt bei 65% (Reviewed in Casey 1998).
