Abstract
The growth declining stage (phase III) of human embryonic fibroblasts (1), is characterized by marked changes in the kinetics of the cell division cycle (2). This is already apparent in the growth curves obtained from daily cell counts performed between the time of subcultivation and the time growth stops (2). The growth curves show that phase III cells have a longer lag phase (time between inoculum and the first net increase in the population), a shorter period of active growth and a saturation density which is considerably less than the one found in early passage cultures (phase II). These changes in growth are due to a longer generation time caused by a prolongation of the G1 and G2 periods of the cell cycle, to a decrease in the amount of cells engaged in the division cycle and to an increase of the nondividing population between subcultivation and the time growth stops.
