Abstract

The immune system, like government, requires a host of checks and balances for normal function. Regulation of immunological responses, including both cell-mediated and humoral activities, involves a complex web of cellular interactions recently described by Jerne (1974) as a “network.” These interactions include a number of stop-and-go signals mediated by various leukocytes and their circulating soluble products. Specificity of these regulatory activities ranges from highly restricted to nonspecific and may or may not include requirements for genetic identity between the regulator and effector mechanisms. Recent efforts have resulted in the characterization of many of these regulatory cells on the basis of membrane antigens or other surface markers (Cantor and Boyse, 1977). An understanding of the molecular mechanisms underlying these immunoregulatory phenomena is also presently emerging, and numerous investigators are attempting to isolate and biochemically characterize soluble immunoregulatory factors. In this chapter, we shall pay particular regard to role of natural immunological control mechanisms and their dysfunction in the pathogenesis of several disease states in man.
