Cyclic AMP Released by Rat Testis during Gonadotrophin Stimulation in vitro

Abstract

THE stimulation of testosterone secretion from testis interstitial cells by luteinizing hormone (LH) and human chorionic gonadotrophin (hCG) is believed to depend on cyclic AMP formation, as is the case with other steroidogenic tissues1. Addition of cyclic AMP to slices of rat2 and rabbit testis3 stimulates testosterone production, and LH increases adenylate cyclase activity in the testis of dog4 and rat5. We have observed that the isolated rat testis is extremely sensitive to LH and hCG stimulation in vitro, responding to low levels of hCG (1 ng/ml.) with maximum release of testosterone into the incubation medium6. In contrast, the teased rat testis responds poorly to gonadotrophic stimulation, and adenylate cyclase in testis homogenates is relatively insensitive to large doses of LH or hCG7.
