Development of a PCR Diagnostic System for Detecting Andean Potato Mottle Virus Associated with Potato Quarantine in Korea

Abstract

One of the four major crops worldwide is the potato. Despite the production of potatoes in Korea, most of them are being imported from other countries. The 15 viral diseases are monitored and tested whenever potatoes are imported in Korea. In this study, there are 2 sets of RT-PCR primer for diagnosing the Andean potato mottle virus (APMoV), which is a type of monitored quarantine viruses infecting potatoes, wherein each nested PCR and modified-positive control plasmid that can verify laboratory contamination and false positive response were developed. The RT-PCR primer sets for analyzing the APMoV were 315 and 732 bp amplified, and each nested PCR were 128 and 391 bp amplified. In addition, the modified-positive control plasmid, which can be used for the PCR diagnosis system, was also developed in order to realize a precision test system for fast and accurate diagnoses of APMoV in imported potatoes. The APMoV PCR diagnosis system, which has been developed in this study, can be used for a possible quarantine in the future. It can also contribute to the increase in the potato production worldwide.

Resumen

La papa es uno de los principales cuatro cultivos a nivel mundial. A pesar de la producción de papa en Corea, la mayoría se importa de otros países. Se monitorean las 15 enfermedades virales y se prueban en cualquier momento en el que se importan las papas. En este estudio, hay dos juegos de iniciadores de RT-PCR para el diagnóstico del virus del moteado de la papa de los Andes (APMoV), que es un tipo de monitoreo de cuarentena de virus que infectan a la papa, donde se desarrollaron cada una de las PCR anidadas y un plásmido testigo modificado positivo que puede identificar contaminación del laboratorio y la respuesta de un falso positivo. Los iniciadores RT-PCR para el análisis de APMoV fueron amplificados en 315 y 732 pb, y cada PCR anidada se amplificó en 128 y 391 pb. Además, el plásmido testigo modificado positivo, que puede usarse para el sistema de diagnóstico de PCR, también se desarrolló para conformar un sistema preciso de prueba para diagnóstico rápido y certero de APMoV en papas importadas. El sistema de diagnóstico de PCR para APMoV que se ha desarrollado en este estudio, puede usarse para una posible cuarentena a futuro. También puede contribuir al aumento en la producción de papa en el mundo.
