Zusammenfassung
In dieser Studie wurde die pharmakodynamische Interaktion zwischen Lumefantrin und Monodesbutyl-Benflumetol bei 44 frischen Isolaten von Patienten mit Plasmodium falciparum Infektionen aus der Gegend von Mae Sot (Thailand) untersucht. Beide Substanzen haben sich als effektiv zur Unterbindung der Parasitenreifung erwiesen, wobei dies bei Monodesbutyl-Benflumetol bei niedrigeren Konzentrationen der Fall war. Der Synergismus der beiden Substanzen wurde bei einer Kombination von Lumefantrin und Monodesbutyl-Benflumetol bei einem Verhältnis von 4,25:1 untersucht. Der geometrische Mittelwert für die vollständige Inhibition der Schizontenreifung lag bei 1035,7 nM für Lumefantrin, 655 nM für Monodesbutyl-Benflumetol und 222,5 nM für die Kombination beider. Eine Interaktionsanalyse wurde gemäß der Methode nach Berenbaum durchgeführt, diese hat einen moderaten Synergismus bei der IC50 ergeben, der bei höheren IC-Werten zunimmt und bei der IC99 im Bereich von ausgeprägtem Synergismus liegt. Der geometrische Mittelwert der Summen der FIC50 beträgt 0,73, bei der FIC90 0,37 und bei der FIC99 0,25.
Summary
The pharmacodynamic interaction between lumefantrine and monodesbutyl-benflumetol has been investigated in 44 fresh isolates of patients with a Plasmodium falciparum infection from the region of Mae Sot (Thailand). Both substances proved to be effective against parasite maturation within the test concentration range, with monodesbutyl-benflumetol being effective at a lower concentration than lumefantrine. Synergism between the two substances was evaluated with a combination of lumefantrine and monodesbutyl-benflumetol at a ratio of 4.25:1. The geometric mean values for complete inhibition of schizont maturation were 1035.7 nM for lumefantrine, 655 nM for monodesbutyl-benflumetol and 222.5 nM for the combination of both. An analysis for interaction according to the method of Berenbaum indicates a moderate synergism at the IC50, which gets stronger with increasing ICs and reaches the highest level at the IC99. The geometric mean of the sums of the FIC50 is 0.73, of the FIC90 it is 0.37 and of the FIC99 it is 0.25.
