Abstract
In this study, we investigated interspecific variations of apple genotypes using ISSR (Inter-Simple Sequence Repeats) and RAPD (Randomly amplified Polymorphic DNA) technique. Total 19 genotypes of wild grown local varieties were collected from Eastern Black Sea region of Turkey and have been studied for the analysis of ISSR and RAPD profile to examine their relationship. From total 53 RAPD and ISSR primer investigated, 28 could amplify clearly and consistently. All bands obtained from ISSR and RAPDs primer were polymorphic.
The dendrogram realized from the RAPD and ISSR markers grouped the 19 genotypes into two major clusters. Cluster I comprised ‘Bey Apple’, ‘Fatty Apple’, ‘Golden Delicious’, ‘Yellow Belly Apple’, ‘Bride Apple’, ‘Green Belly Apple’, ‘Summer Apple’, ‘Yomra Apple’, ‘Tavşanbaşı’, ‘Black Belly Apple’, ‘Chest Apple’, ‘Local Marble’, ‘English Apple’, ‘Willow Apple’, ‘Arabian Girl’, ‘Yellow Hıdır’ and cluster II contained ‘Starking Delicious’, ‘Mahsusa’, ‘Amasya Marble’. Genetic diversity data from this study will be helpful in using and exploiting the wild genetic material for breeding purposes as well as for further research.
Zusammenfassung
In der vorliegenden Studie untersuchten wir die interspezifischen Variationen von Apfel-Genotypen mit ISSR (Inter-Simple Sequence Repeats) und RAPD (Randomly Amplified Polymorphic DNA). Insgesamt neunzehn Genotypen wild wachsender lokaler Sorten wurden von der östlichen Schwarzmeerregion der Türkei für die Analysen gesammelt, um die Zusammenhänge zwischen ISSR und RAPD-Profilen zu untersuchen. Von insgesamt 53 untersuchten RAPD- und ISSR-Primern, waren 28 eindeutig verstärkt. Alle Gruppen waren polymorph.
Das aus den RAPD- und ISSR-Markern erstellte Dendrogramm, bestand aus 19 Genotypen, die in zwei große Gruppen aufgeteilt waren. Cluster I umfasste die Sorten: ‚Bey Apple‘, ‚Fatty Apple‘, ‚Golden Delicious‘, ‚Yellow Belly Apple‘, ‚Bride Apple‘, ‚Green Belly Apple‘, ‚Summer Apple‘, ‚Yomra Apple‘, ‚Tavşanbaşı‘, ‚Black Belly Apple‘, ‚Chest Apple‘, ‚Local Marble‘, ‚English Apple‘, ‚Willow Apple‘, ‚Arabian Girl‘, ‚Yellow Hıdır‘ und im Cluster II waren die Sorten ‚Starking Delicious‘, ‚Mahsusa‘, ‚Amasya Marble‘ enthalten. Die Daten der genetischen Vielfalt aus dieser Studie können zur Verwendung und Nutzung des wilden Genmaterials für die Zucht sowie für weitere Forschungen hilfreich sein.
