Omentin-1 ameliorates oxidative stress in model of diabetic ophthalmopathy via the promotion of AMPK function

Abstract

Background

Omentin-1 is a new type of adipokines.

Objective

To study that the effects of Omentin-1 on diabetic ophthalmopathy and its mechanism.

Results

Omentin-1 expression level in serum of patients with diabetic ophthalmopathy was reduced. In mice with diabetic ophthalmopathy, Omentin-1 mRNA and protein expressions in ocular tissue were suppressed. Furthermore, Omentin-1 presents oxidative stress and ophthalmopathy in the mice model of diabetes. In vitro model of high glucose-induced ophthalmopathy, Omentin-1 reduced oxidative stress and promoted its cell damage, and si-Omentin-1 promoted oxidative stress and protect cells activity. Omentin-1 induced p-AMPK protein expression and reduced ubiquitination of AMPK levels. AMPK inhibitor reversed the function of Omentin-1 on oxidative stress and cells activity in vitro model of high glucose-induced ophthalmopathy. On the other hand, AMPK agonist also reduced the function of si-Omentin-1 on oxidative stress and cells activity.

Conclusion

These findings indicate that Omentin-1 Ameliorates oxidative stress in the model of diabetic ophthalmopathy via the promotion of AMPK function.
