Zusammenfassung
□Hintergrund: In allen Organismen, in denen Sauerstoff verstoffwechselt wird, treten Sauerstoffradikale bzw. reaktive Sauerstoffspezies (ROS) auf, deren Reaktionsfähigkeit durch Radikalvernichtungs- bzw. Scavengersysteme gehemmt wird. Zur Untersuchung dieser Scavengersysteme werden die Aktivitäten von Glutathionperoxydase (GSH-Px) und Superoxiddismutase (SOD) sowie die Konzentrationen von Glutathion (GSH) und Selen bestimmt.
Da in der Literatur sehr untershiedliche Angaben zu diesen Parametern existieren, wäre eine Standardisierung der Bestimmungsmethoden von großem Nutzen.
□Material und Methodik: Erythrozyten von 101 Blutspendern wurden auf die Parameter GSH-Px, GSH, SOD und Selen geprüft. Selen wurde mit der AAS, die GSH-Px-Aktivität nach einer Methode von Paglia et al., GSH nach Adams et al. und SOD mit dem Test Ransod SD 125 bestimmt.
□Ergebnisse: In den Erythrozyten von 101 Blutspendern wurden Selengehalte von 67,1±20,1 nmol/mmol Hb, GSH-Px-Aktivitäten von 842±290 U/mmolHb, GSH von 108±48 µ;mol/mmolHb und SOD-Aktivitäten von 15,8±6,4 U/µ;mol Hb gemessen.
□Schlußfolgerung: Selen, GSH-Px, GSH und SOD in Erythrozyten von Blutspendern sind normal verteilt und zeigen keine signifikanten Unterschiede zwischen weiblichen und männlichen Probanden.
Da keine Standardisierung der Bestimungsmethoden vorliegt, müssen „eigene Referenzbereiche“ verwendet werden.
Abstract
□Background: The oxidant stress is characterized by measurement of the activities of glutathione peroxidase, superoxiddismutase and also by concentrations of glutathione and selenium in erythrocytes.
A standardization of the methods of determination is very important.
□Material and Methods: In erythrocytes of blood donors (n=101) the parameters glutathione peroxidase, glutathione, superoxiddismutase and selenium were determined.
□Results: The following results of the antioxidant parameters in erythrocytes of blood donors were found: Selenium 67.1±20.1 nmol/mmol Hb, glutathione peroxidase 842±290 U/mmol Hb, glutathione 108±48 µ;mol/mmol Hb, superoxiddismutase 15.8±6.4 U/µ;mol Hb.
□Conclusion: Selenium, glutathione peroxidase, glutathione and superoxiddismutase in erythrocytes of blood donors are normally distributed. There are no significant diferences between men and women. The use of “own reference values” is necessary because no standardization of the methods of determination exists.
