The Art of Therapeutic Antibody Discovery: Finding Them One Cell at a Time

Abstract

The immune system is comprised of a complex heterogeneity of cells with dynamic interconnectivity eliciting a diverse array of immunological responses. An individual progeny that has originated from a single clone is programmed to function and behave differently than progenitor or sister cells. Bulk analysis can provide overarching patterns in such a complex system but fails to capture rare subsets and accurately characterize the true individuality of each cell. In the past decade, significant progress in single cell analysis has contributed to the discovery of broadly and highly potent neutralizing antibodies against infectious agents. The discovery of these unique and rare antibody-producing B cells is remarkably valuable for disease prevention, intervention, and vaccine design by mapping the antigenic epitopes. The discovery of these antibodies via single-cell technologies has led to promising diagnostic, treatment, and vaccine development for both viral infection and autoimmune diseases. In this review, we will explore the current advances in antibody discovery using single-cell analysis. We will discuss the major clinical and immunological impacts of antibody discovery for detection, diagnosis, and treatment of infectious diseases and autoimmunity due to the advancement of single-cell technologies.
