FormalPara Synthese – Verteilung – Abbau – EliminationMenschliche Biotinidase ist ein monomeres Glykoprotein mit einer Molmasse von 76 kDa. Es wird in der Leber synthetisiert, und bisher sind 9 Isoformen bekannt. Biotinidase kommt in allen Geweben vor; die höchste Aktivität wurde in Niere, Leber und Nebennieren sowie im Plasma gefunden.
FormalPara PathophysiologieBiotinidase ist das einzige Enzym, das Biotin freisetzten kann. Biotin ist wiederum ein Kofaktor für verschiedene Karboxylasen. Biotinidasemangel führt zu einer schweren, lebensbedrohlichen Erkrankung im Neugeborenenalter bzw. Entwicklungsretardierung bei älteren Kindern. Biotinidasemangel ist behandelbar und die Bestimmung Bestandteil des Neugeborenenscreenings.

FormalPara ProbenstabilitätBiotinidase ist sehr instabil. Plasma bzw. Serum muss rasch gewonnen und sofort bei −70 °C tiefgefroren werden. Versand auf Trockeneis.
