Definition

DARC or Duffy antigen receptor for chemokines is a seven-transmembrane protein, which has a molecular weight of ∼45 k Da. DARC is expressed on vascular endothelium (plasma membrane and caveolae) as well as on red blood cells. It binds to chemokines of both C–C and C–X–C motif chemokines, but signal transduction through the G-protein-coupled receptor or Ca2+ flux is not observed. The genomic organization of the DARC gene consists of two alleles Fya and Fyb that are associated with a polymorphism at the 44-amino acid residue.

Characteristics

DARC is a receptor for malaria parasite Plasmodium vivax, and 70 % of the West African population that lack DARC expression on their erythrocytes are resistant to malaria infection. A novel function of DARC has been identified where it has been found to interact with the metastasis suppressor KAI1CD82.
