Short Description

The aim of the lymphocyte transformation test is to determine by in vitro methods whether a patient has developed a T-cell response against a certain drug. Such T-cell responses are present in practically all types of allergic reactions (type I–IV reactions), which means also in antibody-mediated reactions (Pichler and Tilch 2004). For that purpose, peripheral blood mononuclear cells obtained after density gradient centrifugation (Ficoll-Hypaque) are cultured with different concentrations of the incriminated drug for 5–6 days (Pichler and Tilch 2004; Nyfeler and Pichler 1997; Neukomm et al. 2001; Maria and Victorino 1997; Tsutsui et al. 1992; Naisbitt et al. 2003a, b).
