An enzyme encoded by the POLG gene that is necessary for mitochondrial DNA replication and biogenesis in humans. POLG mutations are associated with mitochondrial diseases; Alpers’ disease, ataxia-neuropathy disorders, dominant and recessive forms of progressive external ophthalmoplegia.
