Amyotrophic Lateral Sclerosis-Related Gene Interactions with Fat Mass and Obesity-Associated Gene

Abstract

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder, characterized by rapid progressive paralysis and death within a few years of the onset of symptoms. Both genetic and environmental factors contribute to the pathogenesis of ALS. Genetic factors can either predispose the onset of the disease or affect its clinical expression. There are several genes linked with ALS, such as SOD1, TARDBP, FUS, C9ORF72, ALS2, SETX, SPG11, VAPB, ANG, and OPTN. A recent study implicated fat mass and obesity-associated (FTO) gene with the pathogenesis of the disease. This warranted further investigation. Therefore, a list of genes associated with ALS was mined from Ensembl and FTO was added on this list to examine its connections with the already established ALS-related genes. In addition, gene ontology analysis was performed to study the relation between FTO and biological, molecular, and cellular aspects of ALS. The results provided some promising connections between FTO and ALS-related genes as well as processes involved in ALS pathogenesis.
