Interaction partners of CDC73 dictate its cellular roles. (a) CDC73 interacts with c-MYC, associating c-MYC with the broader PAF1 complex and leading to inhibition of c-MYC target genes. (b) CDC73 is a member of the PAF1 complex, associating with RNA polymerase II and facilitating transcriptional initiation and elongation. (c) CDC73 is a binding partner of both RNF20 and RNF40, E3 RING finger ubiquitin ligases for histone H2B monoubiquitination at lysine 120 (H2Bub1).
