An Overlay Assay for Detecting Protein Kinase C-Binding Proteins and Substrates

Abstract

Several different approaches have been used to identify protein kinase C (PKC) protein-binding partners. It should be noted that each method may be biased toward identifying a certain type of binding protein, possibly because the methods used emphasize different aspects of PKC-binding protein interactions. This chapter describes features of the overlay assay for identifying PKC-binding partners and briefly compares and contrasts this method with others. Because of the abundance of PKC-binding proteins (1), those cases in which functional relevance has been established are emphasized.
