Abstract

Hematopoietic culture is a rapidly growing area of mammalian cell culture with broad applications. As discussed in Chapter 17, many different types of cells can be produced of multiple lineages and multiple states of differentiation within each lineage. In this chapter, we illustrate the basic techniques and principles of hematopoietic cell culture by providing protocols for hematopoietic cultures of three types: retroviral transduction of CD34+ cells for gene therapy, and expansion of colony-forming unit-granulocyte monocyte (CFU-GM) progenitors and neutrophil postprogenitors for transplantation therapies.
