Reagent and endoproteinase dilutions are calculated based on a final protein concentration of 1 ug/uL (100 ug of protein in 100 uL of solution). This is achieved by mathematically dividing 100 ug (83 uL is the total volume substracted by reagent volumes) by the protein concentration determined in Subheading 3.3. If the concentrated protein is diluted to less than 1.2 ug/uL, adjust reagent and/or endoproteinase volumes to final concentrations stated in the protocol.
