Abstract

Gloriosa superba is a climbing vine belonging to the family Colchicaceae and listed as an endangered species. Seeds and tubers are of medicinal importance which is widely used by the tribal communities. Peculiar structure of the flower hinders the reproduction thereby its regeneration. Artificial pollination is a must which renders the need for pollen collection and thereby evaluating the viability and fertility status of pollen grains. In vitro viability studies were carried out by hanging drop technique using Brewbaker and Kwack’s medium with 10% sucrose solution. Cryopreservation of pollen gains was carried out for long-term storage of germplasm and further breeding programs. Post-cryopreservation viability was also assessed to compare the germination profiles of fresh pollen and cryopreserved pollen grains. This chapter gives a detailed procedure about the pollen cryopreservation protocol in this species right from pollen collection, germination, pollen cryopreservation, and fertility experimenting. The protocol is simple and effective for supplementing to field germplasm collections as well as hybridization programs.
