There are many reasons why it may be desirable to obtain the sequence of a piece of DNA At the highest level, there are currently many genome-sequencing projects underway, the object of which is to obtam the complete sequence of the DNA of a particular organism, human (1), yeast (2) and Cuenorhabdztzs elegans (3) are some examples The hope is that by obtaming the complete sequence, we will gam a much better understanding of how gene expression is controlled and hence a greater understanding of the genetic basis of many diseases (4) Alongside the genome projects, many workers are focusing in on particular genes and at the method of action and control Eventually, with the mtroduction of simpler automated systems for sequencing, It may be possible to perform diagnosis of genetic disease by sequencing All of the areas where sequence mformation is required mean that sequencing is one of the most important technologies currently available to the molecular Biologist.
