Isolation of Total RNA from Plants

About 90% of viruses-infected plants have ssRNA as their genome. Complementary DNA (cDNA) copy for the ssRNA is made using reverse transcriptase enzyme which can then be used as template for PCR assay. This procedure requires isolation of RNA from either purified virus particles or from virus-infected plants. A large number of protocols are available for the extraction of total RNA from different plant species depending on, the type of tissue used for extraction and the presence of inhibitors and secondary metabolites. Majority of the protocols are based on the use of trizol while phenol-based method is the best suitable for species with high amount of secondary metabolites, and use of polyethylene glycol (PEG) may be suitable to extract RNA from species with high amount of polysaccharides and polyphenols.
