Zusammenfassung.
Maligne Tumoren sind zur Invasion in normales Gewebe sowie zur Absiedlung von Metastasen in der Lage. Diese beiden Hauptfaktoren machen die Mortalität und Morbidität von Kolonkarzinomzellen aus. Invasion und Metastasierung erfordern ein Attachment von Tumorzellen an die Basalmembran, die Degradation des lokalen Bindegewebes sowie die Penetration und Migration durch proteolysiertes Stroma. Matrix-Metalloproteinasen (MMPs) sind die Schlüsselenzyme, die für den Abbau der extrazelllulären Matrix (ECM) verantwortlich sind. Bis heute wurden 26 humane MMP-Gene identifiziert, und viele von ihnen sind von onkologischer Relevanz. Die ECM-Degradation durch MMPs erhöht nicht nur die Tumorinvasion, sondern beeinflusst auch das Tumorzellverhalten und führt so zur Tumorprogression. Diese Übersicht berücksichtigt die neuesten zellulären und molekularen Mechanismen der MMPs im Hinblick auf Wachstum, Invasion und Metastasierung von kolorektalen Karzinomen.
Abstract.
Malignant tumors have the ability to invade normal tissue and to spread to distant sites giving rise to metastasis, which are major factors in the morbidity and mortality of colorectal cancer. Invasion and metastasis involve attachment of tumor cells to the basement membrane, degradation of the local connective tissue, and penetration and migration through proteolyzed stroma. The key enzymes responsible for ECM breakdown are matrix metalloproteinases (MMPs). To date, 26 MMP genes have been identified in humans, and many are implicated in cancer. ECM degradation by MMPs not only enhances tumor invasion, but also affects tumor cell behavior and leads to cancer progression. This review highlights recent developments with regard to the cellular and molecular mechanisms of MMPs that influence colorectal tumor cell growth, invasion, and metastasis.
