Zusammenfassung
Die schnelle Entwicklung der Nanotechnologie, der Schlüsseltechnologie des 21. Jahrhunderts, erfordert eine adäquate Risikobetrachtung und Abschätzung von Gesundheitseffekten bei Exposition gegenüber Nanomaterialien. Da die miniaturisierten Stoffe teilweise andere physikalische, chemische (und biologische) Eigenschaften als das Ausgangsmaterial aufzeigen, sollten die für das bulk Material etablierten toxikologischen Erkenntnisse nur bedingt und mit Vorsicht auf Nanoobjekte übertragen werden.
Nach dem derzeitigen Kenntnisstand geht das größte gesundheitliche Risiko von inhalierbaren, nicht oder schwer löslichen Nanomaterialien aus. Diskutiert werden u.a. Wirkungen im Respirationstrakt wie Entzündungsreaktionen, oxidativer Stress und Lungenfibrosen, adverse Effekte im kardiovaskulären System und ZNS. Zudem bleibt eine kanzerogene Wirkung von Nanoröhren, die strukturelle Ähnlichkeit mit Asbestfasern aufweisen, zu befürchten.
Unter Berücksichtigung vielfältiger z.T. bekannter bzw. vermuteter Wirkmechanismen nanoskalierter Materialien haben wir aktuell ein umfangreiches arbeits- und präventivmedizinisches diagnostisches Programm ausgearbeitet mit Untersuchungen zur individuellen Gesundheit, zu persönlichen Risikofaktoren, zu Expositionen, zu speziellen Wirkmechanismen und zu präklinischen Markern von Organwirkungen, welches wir im Folgenden vorstellen.
Summary
The rapid development of nanotechnology, the key technology of the 21st century, merits an adequate evaluation of possible risk factors. There is an urgent need to calculate the health dangers caused by exposure to nanomaterials.
The miniaturized substances show different physical, chemical (and biological) properties than the bulk material — one should not automatically attribute the known same toxic effects of one to the other.
According to present knowledge, the greatest risk arises from inhalation of these not soluble or only slightly soluble particles. One tries to compare the result of exposure on the respiratory tract as a sort of inflammation, oxidation stress and fibrosis of the lung; one considers also the adverse effects on the cardiovascular system and on the central nervous system.
In addition nanotubes with their structure similar to asbestos- fibres may have an carcinogenic potential.
Taking into consideration the various possible working mechanism of nanomaterials, we have come up with an extensive occupational and preventive medical diagnostic program.
Here we would like to publish our examinations suitable for assessing individual health situations, for estimating risk factors, to exposure estimation, to special effect mechanism and to recognize preclinical signs of organ involvement.
