Somministrate i.v. nel cane sveglio normale, la glibenclamide e la glisoxepide sono assai più attive della tolbutamide, se l’effetto viene valutato in termini di abbassamento massimo della glicemia. Somministrata p.o., la glibenclamide agisce sulla glicemia meno rapidamente rispetto alla tolbutamide e alla glisoxepide. Lostudio dinamico della secrezione insulinica dimostra che questa ha un inizio più lento, ma si prolunga per un tempo maggiore con la glibenclamide che non con gli altri due farmaci. Nel pancreas isolato e perfuse-di ratto, la glisoxepide e la tolbutamide stimolano la secrezione insulinica più rapidamente della glibenclamide; con quest’ultima sostanza, la secrezione insulinica si protrae più a lungo e la quantità di ormone liberata è maggiore. Per quanto riguarda l’azione beta-citotrofica, i tre sulfamidici stimolano la neoformazione delle isole di Langerhans nel topo svizzero.

Administrées par voie i.v. chez le chien éveillé normal, la glibenclamide et la glisoxepide sont beaucoup plus actives que la tolbutamide, si l’effet est évalué en termes de réduction maximale de la glycémie. Administrée p.o., la glibenclamide agit sur la glycémie moins rapidement par rapport à la tolbutamide et à la glisoxepide. L’étude dynamique de la sécrétion insulinique montre que celle-ci a un début plus lent mais se prolonge pour un temps plus long avec la glibenclamide qu’avec les deux autres médicaments. Dans le pancréas isolé et perfus de rat, la glisoxepide et la tolbutamide stimulent la sécrétion insulinique plus rapidement que la glibenclamide; avec cette substance, la sécrétion insulinique dure plus de temps et la quantité d’hormone libérée est plus grande. En ce qui concerne l’action bêta-cytotrophique les trois sulfamides stimulent la néoformation des îlots de Langerhans chez le rat suisse.

Suministradas i.v. en un perro despierto normal glibenclamida y glisoxepida son mucho más activas que la tolbutamida si el efecto se evalúa en términos de descenso máximo de la glucemia. Suministrada p.o., la glibenclamida actúa sobre la glucemia menos rápidamente que la tolbutamida y que la glisoxepida. Elestudio dinámico de la secreción insulínica demuestra que esta tiene un inicio más lento, pero que se prolonga durante un tiempo mayor con la glibenclamida que con los otros dos productos. En el páncreas aislado y transfundido de rata, la glisoxepida y la tolbutamida estimulan la secreción insulínica más rapidamente que la glibenclamida; con esta última substancia, la secreción insulínica dura más y la cantidad de hormón liberado es mayor. Por lo que a laacción beta-citotrófica se refiere, los sulfamídicos estimulan la neoformación de las islas de Langerhans en el ratón suizo.
Zusammenfassung
Wenn die Wirkung an der maximalen Blutzuckersenkung gemessen wird, sind i.v. beim normalen wachen Hund verabreichtes Glibenclamid und Glisoxepid bedeutend wirksamer als Tolbutamid. Oral verabreicht wirkt Glibenclamid langsamer auf den Blutzucker als Tolbutamid und Glisoxepid. Diedynamische Untersuchung der Insulinsekretion zeigt, dass diese bei Glibenclamid langsamer in Gang kommt, aber länger anhält als bei den beiden anderen Medikamenten. Am isolierten perfundierten Rattenpankreas regen Glisoxepid und Tolbutamid die Insulinsekretion schneller an als Glibenclamid; nach letzterem hält die Insulinsekretion dagegen länger an und erreicht höhere Werte. Was diebeta-zytotrope Wirkung anbelangt, so regen alle drei Sulfonylharnstoffe die Neubildung der Langerhans’schen Inseln bei Schweizermäusen an.
Summary
The authors conclude that administered i.v. in the normal conscious dog, glibenclamide and glisoxepide are considerably more active on glycemia than tolbutamide. Administered p.o., glibenclamide acts less rapidly on the glycemia than tolbutamide and glisoxepide. Thedynamic study of the insulin secretion shows that the secretion of insulin begins more slowly and lasts longer with glibenclamide than with the two other products. On the isolated and perfused rat pancreas, glisoxepide and tolbutamide more rapidly stimulate the secretion of insulin than glibenclamide; with the latter substance the duration of the secretion of insulin is longer and the total quantity of insulin secreted is greater. As far as thebeta-cytotrophic action is concerned, all three sulfonamides stimulate the neogenesis of the islets of Langerhans in Swiss mice.
