Résumé

L’étude immunochimique des corps d’inclusion formés par lesBaculovirus de Lépidoptères est entreprise en vue de différencier ces viroses. Le travail relaté décrit l’influence de différents solutés sur l’extraction des protéines des granules de la granulose dePieris brassicae. L’intérêt de l’emploi d’agents réducteurs tels que l’acide thioglycolique ou le mercaptoéthanol pour la solubilisation des granules est mis en évidence.

Summary

The proteins of inclusion bodies of Baculoviruses (pathogens causing granulosis and nuclear polyhedrosis) are peculiar to insect viruses. The study of their immunochemical and biochemical characteristics should make it possible to establish an accurate differential diagnosis.

The present work is a preliminary study, the objective of which is to determine optimum conditions for the solubilization of inclusion bodies.

The following solutions are used for dissolving granules of the granulosis ofPieris brassicae:

  1. CO3Na2 (0,06-0,25M)+ClNa 0,05M (Bergold’ process, 1963).

  2. Tris 2 M.

  3. Mixtures of tris and urea.

  4. Sodium thioglycolate-bicarbonate (0,06–0,25 M), pH 10,5.

  5. 2-mercaptoethanol 1% in tris 1 M.

The proteins released are analysed by immunodiffusion and starch gel electrophoresis. Two different proteins are revealed: the T protein is extracted when reducing agents are added (thioglycolic acid, 2-mercaptoethanol) in alkaline solutions; the C protein is released in any case.

The use of reducing agents improves the classical process of solubilization of inclusion bodies.
