Zusammenfassung
Kaffee bzw. das darin enthaltene Coffein vermögen, abgesehen von ihren vielfältigen Organwirkungen, auch den Stoffwechsel des Organismus zu steigern. Der damit einhergehende erhöhte Energieumsatz erfordert den vorherigen Abbau des in der Leber und im Muskel gespeicherten Glykogens zu Glukose bzw. den Abbau von Neutralfetten aus dem Fettgewebe in Fettsäuren und Glycerin, Vorgänge, die als Glykogenolyse bzw. Lipolyse bezeichnet werden. NachSutherland andButcher (1960) werden Glykogenolyse und Lipolyse letztlich durch eine Erhöhung von intrazellulärem cycl. 3,5-AMP verursacht. Im Falle einer Zufuhr von Coffein oder anderer Methylxanthine kann eine Erhöhung von cycl. 3,5-AMP auf verschiedene Art hervorgerufen werden. Sie kann einmal auf einer Hemmung der Phosphodiesterase beruhen, so daß durch langsameren Abbau ein höherer Gehalt an cycl. 3,5-AMP resultiert; ferner kann die Erhöhung von cycl. 3,5-AMP auch durch eine für Methylxanthine nachgewiesene Freisetzung von Katecholaminen zustande kommen. In diesem Fall wird über eine Stimulierung der Adenylcyclase vermehrt cycl. 3,5-AMP synthetisiert. Was die Lipolyse angeht, so wird neuerdings vonSchwabe undEbert (1973) eine durch Methylxanthine bedingte Erhöhung von cycl. 3,5-AMP diskutiert, die durch eine kompetitive Hemmung von Adenosin zustande kommen soll. — In welchem Umfange diese Wirkungsmechanismen in vivo von Bedeutung sind, läßt sich zur Zeit noch nicht entscheiden, da in den bisherigen Untersuchungen am Menschen bzw. am Tier nur das Verhalten der Katecholamine selbst im Blut bzw. Harn oder bestimmte davon abhängige Wirkungen, wie z. B. der Sauerstoffverbrauch, ermittelt wurden.
Summary
Coffee and its most important constituent, caffeine, may not only stimulate the function of many organs but also increase the metabolism in the body. These effects require a higher energy production which is mainly obtained from striated muscles by glycogenolysis and from fat tissue by lipolysis.Sutherland andButcher were able to demonstrate that these degradation processes are primarily caused by an increase of cyclic 3,5-AMP. — In this connection caffeine and other methylxanthines are of special interest because these compounds also increase the intracellular amount of cyclic 3,5-AMP. This effect may be caused by an inhibition of phosphodiesterase, a release of catecholamines with resulting stimulation of adenylcyclase or by competitive inhibition of adenosine. At the present time it cannot be said which of these mechanisms primarily is involved in the in vivo effects of caffeine and other methylxanthines.
