Summary
The effects of PGE2 and PGF2α on corpus uteri and cervix strips of guinea pigs were studied in vitro. Myometrium strips developed maximal contraction activity at concentrations of 10−7 M PGE2 and 5 × 10−7 M PGF2α in the organ bath, respectively. As for corpus uteri myometrium strips, longitudinal (extern) and circular (intern) layers of the organ wall showed comparable Prostaglandin-induced contractions. On the other hand, longitudinal and circular strips from cervix uteri tissue showed adverse Prostaglandin effects: PGE2 relaxed the cervix strips by lowering the muscle tone, the contraction frequency and a complete stop of contractions within most experiments lasting for an average time of 15 min. PGF2α induced strong contractions of the same kind like corpus uteri strips did. The results indicate a role of PGE2 in the opening of the cervix, whereas PGF2α possibly plays a role in the involution of the uterus post partum.
Zusammenfassung
Mit in vitro-Versuchen wurde die Wirkung von PGE2 und PGF2α auf das Corpus uteri und die Zervix des Meerschweinchens 3–4 Tage nach dem Werfen getestet. Bei Konzentrationen von 10−7 M PGE2 bzw. 5 × 10−7 M PGF2α im Gewebebad wurden die Myometriumstreifen zu maximaler Kontraktionstätigkeit stimuliert.
An den Segmenten des Corpus uteri zeigten longitudinale (äußere) und zirkuläre (innere) Myometriumschichten für beide Prostaglandine Kontraktionen, die sich lediglich in der Dauer der initialen Tonuserhöhung unterschieden.
Im Gegensatz dazu konnten an Zervixstreifen konträre Effekte dargestellt werden: PGE2 relaxierte die Zervixstreifen unter Verminderung der Kontraktionsfrequenz, dem Absinken des Muskeltonus und in den meisten Fällen einem völligen Sistieren der Kontraktionen über ca. 15 min. PGF2α dagegen kontrahierte alle Zervixstreifen in vergleichbarer Weise wie die Corpus uteri-Streifen.
Die Befunde deuten darauf hin, daß PGE2 eine Rolle bei der Zervixeröffnung spielt und PGF2α moglicherweise die Involution des Uterus post partum begünstigt.
